ABSTRACT: Insect-transmitted viruses cause some of the most damaging and economically important diseases of crop plants, especially in tropical and subtropical regions. The geminiviruses, a group of single-stranded DNA viruses with unique twined icosahedral virus particles, are responsible for many of these diseases. Of particular importance are the geminiviruses transmitted by whiteflies (Bemisia tabaci), which are in the genus Begomovirus. This is the largest genus of plant viruses (in terms of number of species), and some are responsible for many devastating diseases in vegetable and fiber crops throughout the world. Yellow vein mosaic disease of mesta (kenaf, Hibiscus cannabinus L.; and roselle, H. sabdariffa L.) is a new entrant to the disease scenario and it is associated with a novel monopartite Begomovirus. Changes in different biochemical parameters in diseased mesta plants were observed as compared to healthy ones. Isozyme pattern and assays of different enzymes, namely catalase, acid phosphatase, peroxidase, esterase, polyphenol oxidase and superoxide dismutase, revealed lower activities of catalase, acid phosphatase and peroxidase enzymes and enhanced activities of esterase, polyphenol oxidase and superoxide dismutase in diseased plants as compared to healthy ones. Em 1996 e 1997, observaram-se sintomas de viroses causadas por geminivírus transmitidos por mosca branca em plantas de tomateiro (Lycopersicon esculentum) e pimentão (Capsicum annuum) no Submédio do Vale São Francisco, situado nos Estados de Pernambuco e Bahia. De outubro de 1996 a dezembro de 1998, foram coletadas 1.368 amostras foliares de tomateiro e 194 de pimentão, exibindo sintomas similares àqueles causados por geminivírus, em 104 e 16 plantios, respectivamente, de 12 municípios dessa região e em dois municípios vizinhos. Los begomovirus son patógenos de plantas que en las últimas décadas han causado importantes pérdidas a cultivos de interés económico, en regiones tropicales y subtropicales. En Cuba se han identificado nueve begomovirus afectando cultivos de importancia económica como tomate, fríjol, tabaco, pimiento, calabaza, papa, y malezas de diferentes familias botánicas. This note is the first report of Bemisia tabaci (Gennadius) biotype B colonizing passionvine in Brazil. We examined the colonization of nine Passiflora species by a wild B type population under greenhouse conditions. P. amethystina Mikan was the most preferred species for oviposition and colonization, whereas P. suberosa L., P. coriacea Juss. and two commercially cultivated species, P. alata Curtis and P. edulis Sims f. flavicarpa Degener, were mostly uncolonised. P. morifolia Mast., P. cincinnata Mast., P. foetida L. and P. caerulea L. showed intermediate levels of colonization. Such differential colonization might suggest some degree of resistance by certain Passiflora species or oviposition preference by B. tabaci. |

La amplificación por círculo rodante (ACR) constituye una alternativa eficaz y rápida para los estudios de diversidad biológica de geminivirus en diferentes regiones de mundo. El uso de esta tecnología permite obtener amplificaciones de ADN circulares pequeños y de simple cadena, sin tener en consideración sus posibles variaciones secuenciales. Además, la ACR tiene una alta fidelidad de copia y la cantidad de ADN molde empleada para la amplificación es ínfima. Partial nucleotide sequences of five tomato infecting Begomovirus isolates were determined from DNA-A fragments, corresponding to the 5' region of the replication associated protein gene, the intergenic region and the 5' region of the coat protein gene. Isolate DFM shared 95% identity with Tomato mottle leaf curl virus (TMoLCV), isolates 34, PA-05, and Ta4 were 88% identical to Tomato yellow vein streak virus and isolate DF-BR3 shared 77% identity with TMoLCV. Recombination analysis indicated that isolate DF-BR3 was a chimaera, and it provided evidence that there is a complex and actively recombining population of tomato infecting begomoviruses in Brazil. |

O objetivo deste trabalho foi identificar progênies de feijoeiro (Phaseolus vulgaris) na geração F4 provenientes do cruzamento entre cultivares resistentes ao Bean golden mosaic virus (BGMV). Os parentais foram as cultivares Carioca-MG (suscetível) e IAPAR-57, IAPAR-72 e IAPAR-65 (resistentes). A população de 480 progênies foi obtida pelo modelo dialélico completo. Estas foram semeadas em campo sob inoculação natural e estudada ao nível de plantas individuais para cálculo do índice de doença de BGMV (I.D.) The study aimed to assess the reaction to Tomato yellow vein streak virus (ToYVSV) of determinate-growth tomato breeding lines of the Instituto Agronomico de Campinas (Campinas Agronomic Institute) germplasm collection and to select resistant plants. The biological transmission of Tomato Venezuela virus (ToVEV) by biotype B of the whitefly species Bemisia tabaci (Gennadius) increased (21.7-95.0%), and the time for symptom expression decreased (16-12.6 days) as the number of viruliferous whiteflies allowed access for inoculation to susceptible tomato plants increased from 1 to 20 adults/plant. When acquired only as a nymph, adults of this biotype transmitted the virus to 88.3% of susceptible tomato plants, using 15 viruliferous individuals per test plant, corroborating the circulative nature of the transmission.
